The presence of the small heat shock protein αB-crystallin in ballooned neurons, characteristic of Pick’s and Creutzfeld-Jacob disease, and in Rosenthal fibers, characteristic of Alexander’s disease, suggests a role in the aggregation and remodelling of neurofilaments in neurodegenerative disease. Both mono- and poly-ubiquitinylated conjugates of αB-crystallin have been observed in Rosenthal fibers. Phosphorylated αB-crystallin interacts with FBX4, an F-box-containing protein that is a component of the ubiquitin-protein isopeptide ligase SCF (Skp1/Cul1/F-box), implicating αB-crystallin in the ubiquitin-proteasome pathway.
