Immunogen: Human placenta derived proteasomes.
Specificity: Recognizes human and rabbit proteasome 20S (α6 subunit). Does not cross-react with rat or mouse protein.
Application: Immunohistochemistry
Immunoprecipitation: This antibody has been immobilised on CNBr-activated Sepharose CL-4B and used to precipitate proteasomes from HeLa cells. Proteasomal subunits were subsequently separated by 2D PAGE.
Western Blot: Single dimension SDS-PAGE of a human placental proteasome preparation or HeLa cell lysate followed by Western blotting gives a single band with a relative molecular weight of approximately 33kDa. The pattern seen in the figure has been observed with a large number of antibodies to this subunit. Taken with the fact that no antibody has been identified reactive with only one of these proteins, it is likely that the minor spot with a lower pI is structurally closely related to α6. Proteasome preparations containing degradation products, particularly of subunit α6, often give reaction patterns with several more spots than that shown.

Use/Stability: Aliquot undiluted antibody into smaller volumes (not less than 10µL) prior to freezing if appropriate. The use of high quality ‘antiserum-grade’ plastic or glass vials is recommended. Store diluted antibody at 2-4°C (do not freeze) and use within 1 month. Dilute to working strength with phosphate buffered saline pH 7.2-7.4 and 1% normal goat serum (if a goat anti-mouse IgG linker antibody is to be used). 
Handling: Avoid freeze/thaw cycles.
