非监督的MS/MS二级谱图的相似性代谢轮廓图分析显示与防御有关的土豆代谢物

Solanumtuberosum(potato)wildtypeandoverexpressionlineswithanenhancedPhytophtorainfestansresistancewereanalysedfortheirmetabolicresponsetosimulatedherbivory.• UsingthedataacquiredwithBrukerLC-ESI-Q-TOFMSandMS/MSinstrumentationandmethods,theMetaboScapesoftwareprovidedanefficientplatformforthestraightforwardlinkageofstructuralsimilaritiesandmetaboliteabundances.Weusedvariousstatisticaltools(PCA,t-test)inMetaboScapetofocusonrelevantfeatures.TheunsupervisedMS/MSsimilaritymetaboliteprofilingapproachenabledarapidannotationof17metaboliteswhichwerepreviouslyunknowninouranalyses.• ThecommonabundancepatternstogetherwithcommonMS/MSpatternspointedtoanoverflowofthehydroxycinnamicacidamidebiosynthesisintheAtACT-AtDTX18overexpressionlines.ThisisinperfectaccordancewiththefunctionoftheoverexpressedenzymeAtACTthatisresponsibleforthebiosynthesisoffourHCAAs.TheincreaseofthemanyotherHCAAsmightbeametabolicsideeffectoftheplanttocompensateforthisoverflowandtobypasstoxiceffectsforthecell.MetaboScapewasveryvaluableforeasydataprocessing,theannotationofmanyformerlyunknowncompoundsandanintuitivevisualizationofmetabolicchanges.
