3D可视化和3D图像分析:
MetaXpress软件可以将Z轴序列图像重构成3维立体结构观察细胞核及细胞网络,并进行3D水平的分析。图像应用用户自定义的3D分析中“findfibers”功能,“fibers”数值评价神经突触;同时计算了细胞个数。计算对象首先在每层中识别出,然后使用“connect
bybest
match”功能将其在3D空间中连接起来。3D分析使突触计算更准确,是一个评估神经连接等更好的方法。3D分析得到的参数包括突触个数、细胞体积、总突触体积、分支个数和节点数。细胞总个数是通过在Hoechst染色通道识别“sphericalb
j e c t s ” 。图5
所示为分析识别“fibers”和“nuclei”的蒙板。另一个用户自定义模块分析细胞活力特征(钙黄绿素阳性的胞体)和线粒体完整性(MitoTracker
Orange阳性的胞体)。细胞分类分析可以得到活细胞个数(钙黄绿素阳性的胞体)和有完整线粒体的细胞个数(MitoTracker
Orange阳性)。计算结果包括细胞个数(total nuclei),活细胞个数(Calcein AM
positive),以及细胞体积和荧光强度。图5B和5C所示为“Calcein AMpositive
cytoplasm”,“MitoTracker positive cytoplasm” 和
“nuclei”的识别蒙板。图6显示了化合物影响神经网络的几个特征参数: 突触个数的减少(fibers),节点个数,以及活细胞个数和完
整线粒体的细胞个数。
应用OrganoPlates做神经毒性的3D分析:
表型分析包括在程度和复杂性上定量3D分析神经网络,得到多个参数。在这个神经模型系统里,我们评估了模型的重复性、多参数,并检测了多个已知的神经毒性化合物。通过这些分析方法,我们准确得到了化合物对复杂神经网络的浓度依赖的抑制作用曲线。因此验证了此模型可以用于高通量的多参数的预测化合物的神经毒性。
结论:
我们建立了一个可定量的共聚焦高内涵成像方法, 结合使用M I M E T A SOrganoPlates,对化合物影响神经细胞活力和形状进行高通量的3D表型分析。共聚焦成像和多参数3D分析模块提供了一个在3D水平上计算和统计神经细胞个数和突触特征的方法。3D分析结果可计算出化合物对神经细胞影响的EC50值,帮助筛选出毒性化合物。
