Description
The 5-AMP-activated protein kinase (AMPK), a member of the SNF1 (sucrose nonfermentor) kinase family (1) AMPK is a heterotrimeric protein comprising α (63 kDa), β (38 kDa) and γ (38 kDa) subunits. The alpha subunit is the catalytic subunit (which requires phosphorylation to be active) (2-3). Beta and gamma are noncatalytic subunits, although they have been found to interact with the active subunit in liver. AMPK regulates fatty acid and sterol synthesis by phosphorylation of acetyl-CoA as well as cholesterol synthesis via phosphorylation and inactivation of hydroxymethylglutaryl-CoA reductase (4). AMPK also appears to play a role in protecting cells from stresses that cause ATP depletion by switching off ATP-consuming biosynthetic pathways.

Western blot analysis on 293T cell lysates using anti-Phospho-AMPK alpha 1 (pS487) RabMAb (cat. #2802-1). Cells were either (A) untreated (B) treated with LP.
Immunofluorescent staining of HeLa cells using Phospho-specific anti-AMPK RabMAb (cat. # 2802-1).

Specificity
A phospho specific peptide corresponding to residues surrounding Serine 487 of human AMPK alpha 1 was used as an immunogen.

Storage Condition and Buffer
Store at -20 °C. Buffer: 50 mM Tris-Glycine (pH 7.4), 0.15 M NaCl, 40% Glycerol, 0.01% sodium azide and 0.05% BSA. Stable for 12 months from date of receipt.
