Miscellaneous/General: SUMO proteases (SENPs) contribute to three distinct activities in the cell, processing SUMO precursors to generate mature SUMO, depolymerization of SUMO chains, and deconjugation of SUMO modified proteins. Cleavage occurs after the conserved SUMO Gly-Gly motif either at the scissile peptide bond during processing or at the scissile isopeptide bond during depolymerization and deconjugation There are 6 SUMO specific human SENP enzymes (1-3, 5-7), each of which share between 20-60% sequence identity to each other within their respective catalytic domains. The deSUMOylating enzyme SENP5 is localised primarily in the nucleoli (though a significant amount is present in the cytosol) and has been shown to be essential for cell cycle progression, relocalising to the mitochondrial surface during mitosis, at the G2/M transition prior to nuclear envelope breakdown, resulting in mitochondrial fragmentation. SENP5 shows a preference for deSUMOylation of SUMO-2 and SUMO-3 from substrates relative to SUMO-1, though this isoform specificity may be determined in part by its enhanced cleavage of conjugates linked via specific target protein lysine residue in comparison to others.
