SENP6 exhibits a preference for SUMO-2 and SUMO-3 in deconjugation reactions and SUMO-2/SUMO-3 chains in particular, whereas it processes SUMO precursor proteins poorly. SENP6 depletion leads to SUMO-2 and SUMO-3 accumulation in PML nuclear bodies.

Miscellaneous/General: SUMO proteases (SENPs) contribute to two distinct activities in the cell, processing SUMO precursors to generate mature SUMO and deconjugation of SUMO modified proteins. Cleavage occurs after the conserved SUMO Gly-Gly motif either at the scissile peptide bond during processing or at the scissile isopeptide bond during deconjugation. There are 6 SUMO specific human SENP enzymes (1-3, 5-7), each of which share between 20-60% sequence identity to each other within their respective catalytic domains. In contrast the long N-terminal domains show no sequence similarity and are thought to direct subcellular localization. SENP enzymes have been shown to participate in non-redundant cellular functions such as HIF1α stabilization during hypoxia, mitosis and cytokinesis.
