Description
ATP citrate lyase (ACLY), also known as ATP citrate synthase, is the primary enzyme responsible for the synthesis of cytosolic acetyl-CoA in many tissues. The enzyme is a tetramer of four identical subunits. It catalyzes the formation of acetyl-CoA and oxaloacetate from citrate and CoA with a concomitant hydrolysis of ATP to ADP and phosphate. One of these products, acetyl-CoA, serves several important biosynthetic pathways, including lipogenesis and cholesterogenesis. In nervous tissue, ATP citrate-lyase may be involved in the biosynthesis of acetylcholine. NDPK has been found to phosphorylate ACLY and insulin to increase phosphorylation of ACLY (2).

A. Western blot analysis on 3T3 cell lysates using anti-Phospho- ATP citrate lyase (pT447/pS451) RabMAb (cat. #1914-1). Cells were either (A) untreated (B) treated with insulin.
B. Immunofluorescent staining of HeLa cells using anti-phosho-ATP citrate lyase RabMAb (cat. # 1914-1).

Specificity
A synthetic phospho-peptide corresponding to residues surrounding Thr447 and Ser451 of human ATP-citrate (pro-S) lyase was used as immunogen. The antibody only detects ATP-citrate (pro-S) lyase phosphorylated on Threonine 447 and Serine 451.

Storage Condition and Buffer
Store at -20 °C. Buffer: 50 mM Tris-Glycine (pH 7.4), 0.15 M NaCl, 40% Glycerol, 0.01% sodium azide and 0.05% BSA. Stable for 12 months from date of receipt.
