This International Standard specifies sampling methods for the detection and enumeration of microorganisms on the surface of carcasses or parts of carcasses of slaughtered meat animals. The microbiological sampling can be carried out as part of — process hygiene control (to validate and or verify process control, e.g. total counts and Enterobacteriaceae) in slaughter establishments for large mammals, poultry, and game, — risk-based assurance systems for product safety, and — monitoring or surveillance programmes for the prevalence and/or numbers of pathogenic microorganisms. This International Standard includes the use of excision and swabbing techniques depending on the reason for sample collection. It also includes the use of carcass rinsing for the examination of carcasses of poultry and some small animals. Annex A shows sampling sites on the carcasses of various animal species.

Die vorliegende Technische Spezifikation gilt für die Entnahme von Proben vor der Abgabe im Labor zur mikrobiologischen Untersuchung. Sie stellt allgemeine Anweisungen für und spezifische Anforderungen an die Entnahme von Proben und deren Transport zum Labor bereit. Probenahmepläne fallen nicht in den Anwendungsbereich dieser Technischen Spezifikation. Diese Technische Spezifikation gilt für alle Lebens- und Futtermittel, einschließlich Blöcken von gefrorenen Produkten, Schlachttierkörpern (jedoch mit Ausnahme der Oberflächen-Probenahme von Schlachttierkörpern), Fleisch und losen Produkten (Schüttgütern). Dieser Teil von ISO 16649 legt ein horizontales Verfahren für den Nachweis und die Zählung vonGlucuronidase-positiven Escherichia coli durch Kultivierung im flüssigen Nährmedium und Berechnung der wahrscheinlichsten Keimzahl (en: most probable number, MPN) nach der Bebrütung bei (37 ± 1) °C und anschließend bei (44 ± 1) °C fest. Dieser Teil von ISO 16649 ist anwendbar auf: Erzeugnisse, die für den menschlichen Verzehr und zur Fütterung von Tieren bestimmt sind; Umgebungsproben im Bereich der Herstellung und Handhabung von Lebensmitteln. Das Verfahren ist geeignet für die Zählung von Zellen von E. coli, die möglicherweise Stress ausgesetzt waren, bedingt durch Austrocknung, Gefrieren und dem Ausgesetztsein einer salzhaltigen (z. B. marinen) Umgebung oder Beschädigung durch Desinfektionsmittel, wie z. B. chlorhaltige Produkte. Eine Anwendungsbeschränkung dieses Teiles der ISO 16649 wird durch die Anfälligkeit des Verfahrens für ein hohes Maß an Variabilität bedingt. Das Verfahren und die Auswertung der Ergebnisse sind unter Berücksichtigung der in Abschnitt 11 angeführten Informationen durchzuführen. Dieses Verfahren wurde noch nicht vollständig für alle Matrizes bewertet, z. B. für Milch und Milchprodukte. Für Milch und Milchprodukte ist ISO 7251 vorgesehen.
